The DII-VENUS reporter encodes a chimeric protein composed of a fusion between DII of IAA28 and a nucleus-targeted fluorescent protein (VENUS). The targeted degradation of the DII-containing chimeric protein by auxin results in a loss of fluorescence and thus marks the accumulation of auxin in the cell.
