Optimal cutting temperature (OCT) is a compound especially good for preserving cellular ultrastructure (histology) and likely nucleic acids as well. It is also used for specialized applications where formalin fixed tissue is not optimal. This standard operating procedure (SOP) describes how tissues are frozen in OCT.

2023年5月10日 · Orient the tissue as desired. Freeze the tissue using the isopentane bath. Hold the tissue mold over the isopentane bath with forceps for 10 – 20 seconds until the tissue block turns opaque. Avoid getting any isopentane on the OCT compound. Store the frozen tissue at -80°C until ready for sectioning.
Most frozen tissue samples used for biomedical research are collected and embedded in O.C.T. (Optimal Cutting Temperature) compound. O.C.T. has been known to cause ion suppression and decrease spectrum quality from MS experiments (Schwartz, Reyzer & Caprioli 2003).
